I have done it twice. If I recall correctly, it is around 14-15 hours.

Why don't you ride down?
It is less than 1000 kms to Palermo from Rome and a 20 minute ferry ride from mainland Italy to Messina in Sicily.
Unless you want to see Palermo, you can then go straight from Messina to Pozzalo for your ferry ride to Malta (the ferry ride is a couple of hours).
Just my opinion, but to me the east coast of Sicily is way more interesting (Catania, Syracuse etc.).
