To check for air leakage into the carb manifold, what should I spray onto the manifold area, and what should I be looking for from the engine?

IF the smoke trick won't work for you, try either some brake cleaner or carb cleaner in an aerosol can.
You should hear an increase in rpm if the junction you are spraying is the culprit.

+1 for the carb cleaner. You should hear the engine 'smooth out' and the idle speed will stabilize for a second or two as the liquid forms a momentary seal.
